It begins at Exhibition Place with the Dufferin Gates, travels north to Queen Street West where an underpass is needed using Gladstone Avenue to reconnect to the Street north of Queen. From there it goes straight up through numerous neighbourhoods and into North York, where it is interrupted again by Downsview Park.
Between Wilson Avenue and Sheppard Avenue West, it becomes Allen Road before returning to its Dufferin street name. It continues north and goes past Steeles Avenue West north of the City.
The 29 Dufferin bus travels from The Ex to Wilson Station and picks up and drops off people now and then.
